Detection and characterization of Plasmodium spp. by semi-nested multiplex PCR each in mosquito vectors and in people residing in traditionally endemic areas of Paraguay
In Paraguay, no instances of Malaria have been recorded since 2011. Microscopy and the SnM-PCR approach have been applied to detect and characterize Plasmodium spp. each in mosquitoes and in people residing in traditionally endemic areas of Paraguay, to guage the potential of discovering asymptomatic instances and/or Plasmodium parasites circulating in anophelines.
Between 2013 and 2015, 361 human blood samples have been collected on filter paper, and between 2016 and 2017, 938 feminine Anopheles mosquitoes have been captured in 15 Paraguayan localities. All of the diagnostic methods confirmed adverse outcomes. We noticed no asymptomatic case or Plasmodium circulating in vectors.

Suitability of pepper gentle mottle virus as a human enteric virus surrogate for assessing the efficacy of thermal or free-chlorine disinfection processes by utilizing infectivity assays and enhanced viability PCR
Evaluating the efficacy of disinfection processes to inactivate human enteric viruses is essential for the prevention and management of waterborne illnesses attributable to publicity to these viruses by way of ingesting water. Right here, we evaluated the inactivation of two consultant human enteric viruses (adenovirus kind 40 [AdV] and coxsackievirus B5 [CV]) by thermal or free-chlorine disinfection.
As well as, we in contrast the infectivity discount ratio of a plant virus (pepper gentle mottle virus [PMMoV], a lately proposed novel surrogate for human enteric viruses for the evaluation of virus elimination by coagulation‒fast sand filtration and membrane filtration) with that of the 2 human enteric viruses to evaluate the suitability of PMMoV as a human enteric virus surrogate to be used in thermal and free-chlorine disinfection processes.
Lastly, we examined whether or not standard or enhanced viability polymerase chain response (PCR) evaluation utilizing propidium monoazide (PMA) or improved PMA (PMAxx) with or with out an enhancer might be used as options to infectivity assays (i.e., plaque-forming unit methodology for AdV and CV; native lesion rely assay for PMMoV) for evaluating virus inactivation by disinfection processes. We discovered that PMMoV was extra proof against warmth therapy than AdV and CV, suggesting that PMMoV is a possible surrogate for these two enteric viruses with regard to thermal disinfection processes.
Nonetheless, PMMoV was rather more proof against chlorine therapy in contrast with AdV and CV (which is chlorine-resistant) (CT worth for 4-log10 inactivation: PMMoV, 84.5 mg-Cl2·min/L; CV, 1.15-1.19 mg-Cl2·min/L), suggesting that PMMoV shouldn’t be helpful as a surrogate for these enteric viruses with regard to free-chlorine disinfection processes.
For thermal disinfection, the magnitude of the sign discount noticed with PMAxx-Enhancer-PCR was comparable with the magnitude of discount in infectivity, indicating that PMAxx-Enhancer-PCR is a possible various to infectivity assay.
Nonetheless, for free-chlorine disinfection, the magnitude of the sign discount noticed with PMAxx-Enhancer-PCR was smaller than the magnitude of the discount in infectivity, indicating that PMAxx-Enhancer-PCR underestimated the efficacy of virus inactivation (i.e., overestimated the infectious virus focus) by chlorine therapy.
However, among the many PCR approaches examined within the current research (PCR alone, PMA-PCR or PMAxx-PCR both with or with out enhancer), PMAxx-Enhancer-PCR supplied probably the most correct evaluation of the efficacy of virus inactivation by thermal or free chlorine disinfection processes.
Detection of Leishmania infantum DNA by real-time PCR in saliva of canine
This research developed a real-time quantitative PCR (qPCR) assay to detect L. infantum kinetoplast DNA (kDNA) in canine saliva. The qPCR confirmed an effectivity of 93.8%, a coefficient of correlation of 0.996 and a detection restrict of 0.5 fg/response (0.005 parasites), though it detected till 0.25 fg/response (0.0025 parasites). When samples from 12 canine experimentally contaminated with L.
infantum have been collected, L. infantum kDNA was detected at 16-weeks post-infection (wpi) in 41.7% and 91.7% of saliva and bone marrow samples, respectively, and at 47-wpi in 75% of each samples. L. infantum kDNA might be detected by qPCR in canine saliva, with decrease sensitivity within the early phases of an infection and a decrease parasite load estimation in comparison with bone marrow.
Nonetheless, saliva had comparable sensitivities to bone marrow within the later phases of the an infection and might be used to detect L. infantum kDNA being conscious of its limitations.

T2235004 | Biochain | 5 slides | EUR 308.4 |
Description: Our tissue products are produced by strictly following the IRB ethical standards and procedures and from highest quality tissues. Immediately after collection the tissues are placed in liquid nitrogen and examined by certified pathologists. The thickness of each individual section is ~5um. They are Hematoxylin and Eosin stained and quality tested by immunostaining with anti-beta-actin antibodies. Our tissue products are suitable for various studies on cellular level (RNA localization, Protein expression, etc.) on both normal and pathological cases. It is also an excellent control and educational tool. |
